Effect of 7-day treatment with a novel orally active growth hormone (GH) secretagogue MK-677 on 24-hour GH profiles, insulin-like growth factor-I, and adrenal cortical function in normal young men.


To evaluate the efficacy of a novel analogue of the GH-releasing peptide (MK-677), nine healthy young men participated in a randomized, double-blind, three-period crossover study comparing oral placebo with doses of 5 to 25 mg of MK-677.
Each period involved 7 consecutive days of bedtime administration of the drug.
At the end of each period, plasma levels of insulin-like growth factor-I (IGF-I) and IGF-binding protein-3 (IGFBP-3) were measured at 07:45 h, and 24-h profiles of plasma GH and cortisol were obtained at 15-min intervals along with 24-h urine excretion.
Profiles of plasma free cortisol were calculated at hourly intervals.
Although the amount of GH secreted was similar in all three conditions, GH pulse frequency increased with both doses of the drug, primarily due to an increase in the number of low-amplitude pulses.
Plasma IGF-I levels increased in a dose-dependent manner, whereas IGFBP-3 levels increased only at the highest dose.
There was a positive relationship between GH pulse frequency and IGF-I increase.
Except for the increase in nocturnal waking and morning elevation, MK-677 had no effect on the cortisol profile.
In particular, the 24-hour average plasma total cortisol and free cortisol and urinary excretion of free cortisol were similar under all conditions.
Current data suggest that the use of MK-677 in the treatment of relative body weight deficiency, particularly in adults with impaired immune function due to such deficiencies, warrants further investigation.

And here's a quick note on the effects of cortisol.
Many of these peptides increase cortisol, which is absolutely undesirable during peak GH release if physical conversion is the goal.
Many of these also increase prolactin, which can cause many side effects.
Therefore, an orally active GH releaser that does not increase cortisol would clearly be a game changer.

Two months of treatment with the oral growth hormone (GH) secretor MK-677 in obese subjects increased GH secretion, fat-free mass, and energy expenditure.

We conclude that 2 months of treatment with MK-677 in healthy obese men resulted in sustained increases in serum levels of GH, IGF-I, and IGF-binding protein-3. The effect on cortisol secretion was transient.
Changes in body composition and energy expenditure were anabolic in nature, due to sustained increases in lean muscle mass without fat gain and transient increases in basal metabolic rate.
Further studies are needed to evaluate whether higher doses or longer treatment durations of MK-677 can promote body fat loss.


Another quick point.
Increased IGF-BP3 is a good thing.
In some cases, we see a binding protein, think of SHBG, Sex Hormone Binding Globulin, which definitely reduces the effectiveness of testosterone.
However, in this case, the binding material IGF-BP3 is required to transport IGF-1 into cells and activate the receptor.
Therefore, increasing both IGF-1 and BP3 is a definite benefit.

How to take:
We recommend taking 1-2 capsules daily before bed to reduce appetite and promote relaxation.
It can be taken stacked with L-Dopa and Berberine or various GDAs to eliminate changes in insulin sensitivity and prolactin.
